La perméabilité est un facteur déterminant de l’absorption orale et de la distribution systémique des médicaments. Nos modèles in vitro , tels que PAMPA et les monocouches cellulaires (Caco-2, MDCK-MDR1/BCRP), permettent une évaluation mécanistique de la diffusion passive et du transport dépendant des transporteurs. Ces systèmes permettent une évaluation du transport bidirectionnel et expriment des transporteurs d’efflux humains clés, notamment la glycoprotéine P (MDR1) et BCRP, qui influencent significativement la distribution des médicaments.

La caractérisation des interactions des composés avec ces transporteurs permet de prédire avec précision la pharmacocinétique in vivo et d’évaluer les risques potentiels d’interactions médicamenteuses. Ces connaissances renforcent la modélisation pharmacocinétique, la conception de médicaments pour le SNC et la prise de décisions stratégiques dans les programmes de recherche.

Des scientifiques réalisent des tests de perméabilité en laboratoire pour évaluer le transport des médicaments à travers les barrières biologiques et l'exposition systémique.
