Smart Column Coupling: Overcoming a Three-Way Separation

Challenge :The goal was to separate a starting material and two product isomers, but no single HPLC column could resolve all three.

Approach

Using an X-Select column with a 55–75% ACN/Hβ‚‚O (+FA) gradient, we achieved full separation in a single purification run.

  • Kinetex resolved the isomers but not the SM
  • X-Select resolved the SM but not the isomers
  • Solution: Serially connected both columns

Outcome

  • Full resolution achieved in one injection
  • Reduced time, complexity, and purification steps
  • Pure collection of the two desired isomers
