SfN 2025 – Electrophysiological Characterization of iPSC-Deriver RealDRGx Sensory Neurons
Human dorsal root ganglion (DRG) neurons are central to sensing pain and environmental stimuli but have historically been difficult to study on automated electrophysiology systems. In this study, Sygnature Discovery used the SyncroPatch 384i platform to characterise hiPSC-derived RealDRGx™ sensory neurons, demonstrating their suitability for automated patch-clamp recording. Across six weeks of maturation, the neurons displayed expected voltage- and ligand-gated channel activity, alongside spontaneous and evoked firing. The work confirms the potential of combining iPSC-derived sensory neurons with automated electrophysiology to increase throughput in pain research, supporting hit identification and mechanistic understanding in inflammatory and neuropathic pain drug discovery
