
{"id":17172,"date":"2020-10-22T11:00:58","date_gmt":"2020-10-22T11:00:58","guid":{"rendered":"https:\/\/www.sygnaturediscovery.com\/blog\/renasci-dio-mouse-model-glp-1-semaglitude\/"},"modified":"2020-10-22T11:00:58","modified_gmt":"2020-10-22T11:00:58","slug":"renasci-dio-mouse-model-glp-1-semaglitude","status":"publish","type":"blog","link":"https:\/\/www.sygnaturediscovery.com\/fr\/blog\/renasci-dio-mouse-model-glp-1-semaglitude\/","title":{"rendered":"The dietary-induced obese (DIO) mouse: A robust platform for the development of anti-obesity pharmacotherapies"},"content":{"rendered":"<p>There is a growing need for efficacious\u00a0<a href=\"https:\/\/www.sygnaturediscovery.com\/drug-discovery\/in-vivo-pharmacology\/metabolic-disorder-models\/\"><span style=\"color: #00ccff;\">anti-obesity<\/span><\/a><span style=\"color: #00ccff;\">\u00a0<\/span>treatments that produce clinically meaningful, sustained weight loss and ultimately reduce the risk of developing associated metabolic comorbidities, such as\u00a0<a href=\"\/drug-discovery\/in-vivo-pharmacology\/fibrosis-models\/\"><span style=\"color: #00ccff;\">diabetes<\/span><\/a>,\u00a0<span style=\"color: #00ccff;\"><a style=\"color: #00ccff;\" href=\"\/drug-discovery\/in-vivo-pharmacology\/fibrosis-models\/\">non-alcoholic steatohepatitis (NASH)<\/a><\/span>\u00a0and cardiovascular disease (CVD).<\/p>\n<p>Of particular promise is the array of new pharmacological analogues of the glucagon-like peptide 1 (GLP-1), as well as other incretin-based approaches.<\/p>\n<p>As such, in a continued effort to validate our models with clinically relevant comparators, we recently demonstrated that the long-acting GLP-1 receptor agonist, Semaglutide, was efficacious in our mouse model of dietary induced obesity (DIO).<\/p>\n<p>We showed that twice-weekly administration of Semaglutide produced an impressive\u00a0<strong>24% reduction in body weight<\/strong>, which was associated with a\u00a0<strong>33% reduction in food intake<\/strong>,\u00a0<strong>57% decrease in adiposity<\/strong>\u00a0and\u00a0<strong>improved glucose control<\/strong>. These findings agree with the effects currently being reported in the clinic, and further demonstrate the predictive validity of our DIO mouse model for the development of anti-obesity pharmacotherapies.<\/p>\n<p>\u00a0<\/p>\n<h4><span style=\"color: #006aab;\">Semaglutide reduced body weight by 24% and decreased cumulative food intake by 33% in the DIO mouse model:<\/span><\/h4>\n<p><img decoding=\"async\" class=\"alignnone size-medium wp-image-10651\" src=\"https:\/\/www.sygnaturediscovery.com\/wp-content\/uploads\/2025\/11\/1-1536x558-1-800x291-1.jpg\" alt=\"\"><\/p>\n<p>\u00a0<\/p>\n<h4><span style=\"color: #006aab;\">Semaglutide decreased overall adiposity by 57% in the DIO mouse model:<\/span><\/h4>\n<p><img decoding=\"async\" class=\"alignnone size-medium wp-image-10653\" src=\"https:\/\/www.sygnaturediscovery.com\/wp-content\/uploads\/2025\/11\/2b-1536x530-1-800x276-1.jpg\" alt=\"\"><\/p>\n<p>\u00a0<\/p>\n<h4><span style=\"color: #006aab;\">Semaglutide improved glucose tolerance during an oral glucose challenge in the DIO mouse model:<\/span><\/h4>\n<p><img decoding=\"async\" class=\"alignnone size-medium wp-image-10654\" src=\"https:\/\/www.sygnaturediscovery.com\/wp-content\/uploads\/2025\/11\/3-1536x716-1-800x373-1.jpg\" alt=\"\"><\/p>\n<p>\u00a0<\/p>\n<h4><span style=\"color: #006aab;\">Semaglutide decreased liver lipids (triglycerides, cholesterol, NEFA) in the DIO mouse model:<\/span><\/h4>\n<p><img decoding=\"async\" class=\"alignnone size-medium wp-image-10655\" src=\"https:\/\/www.sygnaturediscovery.com\/wp-content\/uploads\/2025\/11\/4-1536x549-1-800x286-1.jpg\" alt=\"\"><\/p>\n<p>\u00a0<\/p>\n<p>Importantly, our DIO mouse model is generated in-house by feeding a diet closely resembling the fat content of the western diet.
