In the last 5 years or so the interest in psychedelics as therapeutics has been pursued by multiple biotech companies for the treatment of depression, PTSD, substance abuse disorder, and other CNS disorders. Much of the focus has been on known psychedelics such as psilocybin, close analogues, and formulations thereof. However, beyond this, there is an increasing focus on novel chemical entities (NCEs).<\/p>\n<h2>DMPK \u2013 improving on current Psychedelic and close analogues<\/h2>\n<p>One key aim has been to maintain the therapeutic effect of known psychedelics but limit the length of the psychedelic experience, as this brings challenges for real-world treatment adoption. This requires extensive experience and capabilities in DMPK, particularly in how pharmacokinetic properties impact pharmacology. A close collaboration between Chemistry and DMPK \u00a0allows a drug discovery team to generate SAR across a range of analogues of known psychedelics. This is also important in generating novel IP in a highly competitive space.<\/p>\n<h2>5-HT2A Receptor Modulation &amp; Pharmacology \u2013 novel chemical entities and deeper understanding<\/h2>\n<p>Alongside the modulation of DMPK properties, it\u2019s important to consider how analogues of known psychedelics and NCEs engage and modulate the 5-HT2A receptor, as this can also impact their therapeutic and side effect properties. Several approaches to modulating the 5-HT2A receptor that are potentially interesting to generate novel profiles include:<\/p>\n<ul>\n<li>Full agonism vs. partial agonism<\/li>\n<li>Affinity and on\/off kinetics at the receptor<\/li>\n<li>Functional selectivity or biased cell signalling<\/li>\n<li>Orthosteric agonism vs. allosteric modulation<\/li>\n<\/ul>\n<p>To understand drugs with these potential profiles you need:<\/p>\n<ul>\n<li>Cell line engineering capabilities to generate robust, reproducible, and reliable <a href=\"https:\/\/www.sbdrugdiscovery.com\/cell-line-generation.asp\">cell lines<\/a> expressing 5-HT2A receptors and assays measuring G-protein, \u03b2-arrestin, and other effectors<\/li>\n<li>Access to a platform of <a href=\"https:\/\/www.sygnaturediscovery.com\/news-and-events\/blog\/biophysics-how-to-choose-the-right-assay-for-your-project\/\">biophysical assays<\/a> including; SPR, MST, and others, that allow you to use the best assay to answer the question at hand<\/li>\n<\/ul>\n<p>Moreover, it is unclear if engaging the 5-HT2A receptor alone is sufficient to drive a therapeutic response. Molecules with activities at a broader range of 5-HT receptors, or indeed other targets, is an additional poly-pharmacology approach that might be valuable<\/p>\n<h2>Downstream of 5-HT2A receptor modulation \u2013 confidence in therapeutic response<\/h2>\n<p>As a drug discovery project progresses and understanding of <a href=\"https:\/\/www.sygnaturediscovery.com\/drug-discovery\/dmpk-and-physical-sciences\/\">DMPK<\/a> and primary 5-HT2A receptor modulation and pharmacology improves, it becomes clear that secondary, tertiary, and orthogonal assays are necessary to increase confidence, understand downstream effects, and predict therapeutic response. This includes:<\/p>\n<ul>\n<li>Cross-species differences \u2013 using cell lines and primary rodent\/post-mortem human tissue<\/li>\n<li>Endpoints indicative of therapeutic response, mechanism, and side effects (in-vitro, ex-vivo, in-vivo assays), e.g. \u2013\n<ul>\n<li>Neuroplasticity \u2013 neurite outgrowth, molecular marker changes (e.g., egr1\/2, PSD-95, synaptophysin)<\/li>\n<li>IPone accumulation\/Ca<sup>2+<\/sup> response \u2013 measured in-vitro, ex-vivo<\/li>\n<li>Head-twitch response in rodents \u2013 a potential predictor of hallucinogenesis<\/li>\n<li>Microdialysis \u2013 neurochemical profile of your NCE (monoamines, glutamate, GABA)<\/li>\n<li>Drug discrimination \u2013 a sensitive assay of hallucinogenesis and drug abuse<\/li>\n<li>Neuroinflammation (LPS-induced changes in brain cytokines, 1BA1, and TSPO)<\/li>\n<\/ul>\n<\/li>\n<li>PK\/PD, Biomarkers and Translatability\n<ul>\n<li>Ex-vivo binding assays \u2013 e.g. [<sup>3<\/sup>H]-Cimbi-36, [<sup>3<\/sup>H]-MDL100,907, [<sup>3<\/sup>H]-UCB-J<\/li>\n<li>Functional Pharmacodynamic markers \u2013 mydriasis\/miosis, body temperature, endpoints that can be assessed in rodents and early trials in man<\/li>\n<li>Biofluid biomarkers<\/li>\n<\/ul>\n<\/li>\n<\/ul>\n<h2>5-HT2A presentations \u2013 Society for Neuroscience (SfN), Washington, November 11-15, 2023<\/h2>\n<p>At the SfN meeting, Sygnature Discovery and SB Drug Discovery will be showcasing some of the capabilities in the 5-HT2A\/psychedelic space with two presentations.<\/p>\n<p><em>A Cascade of binding and cell-based assays to characterize 5-HT2A receptor ligands and aid the investigation of signalling pathway bias<\/em> <em>(presentation: Gerrard et al.,)<\/em><\/p>\n<p><em> <img loading=\"lazy\" decoding=\"async\" class=\"wp-image-16055 aligncenter\" src=\"https:\/\/www.sygnaturediscovery.com\/wp-content\/uploads\/2025\/11\/5ht2a-graph-for-blog-.png\" alt=\"\" width=\"547\" height=\"287\"><br>\n<\/em>This poster showcases a cascade of <em>in vitro\u00a0<\/em>binding and cell-based assays measuring calcium mobilisation, \u03b2-arrestin interaction, receptor internalisation, and pERK levels. Psychedelic and non-psychedelic ligands were tested for their effect on these different downstream signalling effectors after binding to the receptor.<\/p>\n<p><span style=\"font-size: 14pt;\"><strong><em>Hear more about this topic at Dr Davide Pau\u2019s poster presentation on Monday, Nov 13<sup>th<\/sup>, 1-5 PM, PSTR310.01<\/em><\/strong><\/span><\/p>\n<p>\u00a0<\/p>\n<p><em>Psilocybin: the relationship between head twitch response, locomotor activity, 5-HT<sub>2A<\/sub> ex vivo receptor occupancy and plasma and brain levels of psilocin <\/em>(Cheetham et al.,)<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\" wp-image-16056 aligncenter\" src=\"https:\/\/www.sygnaturediscovery.com\/wp-content\/uploads\/2025\/11\/figure-for-psychedelic-blog.png\" alt=\"\" width=\"438\" height=\"452\"><\/p>\n<p>This poster showcases our deep emphasis on PK\/PD in all drug discovery projects. We show a correlation between <em>ex vivo<\/em> receptor occupancy, plasma and brain OK of psilocin, and behavioural response. Also, for <em>ex vivo<\/em> binding we used the agonist radioligand [<sup>3<\/sup>H]Cimbi-36 which labels the high affinity active state of the receptor.
